Characterization of M3 Mutants of Seri 82 Bread Wheat Cultivar Using Functional Markers

Özet: In this study, Seri 82 bread wheat and 6 mutant lines (Seri 82-M3-1, Seri 82-M3-2, Seri 82- M3-3, Seri 82-M3- 4, Seri 82-M3-5 and Seri 82-M3-6) were screened for some traits using 17 allele specific DNA markers. In the study Seri 82 bread wheat and 6 mutant lines characterized for alleles gluten strength (Bx7OE marker), yellow rust Yr45 (Xwgp115 marker) and Yr51 (SUN104 marker), stem rust Sr49 (SUN209 and SUN479 markers), vernalisation Vrn-A1 (VRN1 marker), dwarfing Rht-B1a (tall) (BF-WR1 marker), Rht-B1b (short) (BF-MR1 marker), Rht-D1a (tall) (DF2-WR2 marker), Rht-D1b (short) (DF MR2 marker), Rht8 (WMS261 marker), high protein content Gpc-B1 (UHW89 marker), rye translocation (RIS, NOR and SCM9 markers), grain hardness Pina (Pina-D1 marker) and waxy Wx-A1 (SUN1 marker). According to the results, 61 polymorphic bands obtained from 17 DNA markers and the average allele number was 3.58. The highest band numbers were obtained from RIS and SCM9 markers with 6 bands, while the lowest band numbers were obtained from Bx7OE, Xwgp115 and SUN209 markers. The average polymorphism information content (PIC) value was determined as 0.79, while the highest PIC value was obtained from BF-WR1 markers as 0.99. As the allele specific markers evaluated stem rust Sr49 was determined in Seri 82, Seri 82-M3-2, Seri 82-M3-3, Seri 82-M3-4 and Seri 82-M3-6 genotypes while dwarfing gene (short) screened in Seri 82-M3-2 genotype and dwarfing gene (tall) determined on the Seri 82, Seri 82-M3-1, Seri 82-M3-3, Seri 82-M3-4, Seri 82-M3-5 and Seri 82-M3-6 genotypes. Rht8 dwarfing gene was found in Seri 82-M3-1, Seri 82-M3-4 and Seri 82-M3-6 genotypes and rye translocation was found in all mutant lines.
