Özet: The Nested PCR is one of the most popular methods for improving the specificity of the PCR techniques. It is assumed that NestedPCR increase 104 fold for accuracy of the amplification of the target DNA sequence. This reaction mechanism contains two consecutivepolymerase chain reactions. The two specific primer of outer region of DNA are used and the long area is replicated at the firstamplification. The two primers of inner area that is come from first amplification are used for reproduction of small area at the secondamplification step. There is no undesirable DNA sequence after the second PCR step. Nowadays this system have been using for GMO,pathogen diagnosis, viral infections, gene expression, fungal diagnosis and many medical fields.
